TOWARD DETERMINING THE ROLE OF PKA IN CONTROLLING TORC2 FUNCTION AND CHEMOTAXIS IN DICTYOSTELIUM DISCOIDEUM by

چکیده
Chemotaxis is a process whereby singleand multi-cellular organisms migrate in response to external chemical stimuli. This directed cell movement is regulated by complex signaling pathways and is implicated in embryonic development, immune response, and the metastasis of cancer cells. Dictyostelium discoideum, social amoebae with the ability to migrate and aggregate in response to chemoattractants such as cAMP, have been used as a model system to study chemotaxis. Preliminary research suggests that protein kinase (PKA) is involved in some of the signaling pathways that regulate chemotaxis. The role of PKA in chemotaxis was investigated, first, by characterizing the phenotype of PKA null cells using established cell biological and biochemical assays. Furthermore, spatiotemporal regulation of critical cytoskeletal proteins was probed in wild-type and PKA null cells using confocal fluorescence microscopy, indicating misregulation of both F-actin and Myosin II in pkaCand pkaRcells. Finally, preliminary work was done to lay the groundwork for experiments exploring possible PKA targets mediating TORC2 function in chemotaxis.

Phosphoinositide-dependent protein kinase (PDK) activity regulates phosphatidylinositol 3,4,5-trisphosphate-dependent and -independent protein kinase B activation and chemotaxis.
Chemotactic cells must sense shallow extracellular gradients and produce localized intracellular responses. We previously showed that the temporal and spatial activation of two protein kinase B (PKB) homologues, PkbA and PkbR1, in Dictyostelium discoideum by phosphorylation of activation loops (ALs) and hydrophobic motifs had important roles in chemotaxis. We found that hydrophobic motif phosph...
متن کاملPakD, a putative p21-activated protein kinase in Dictyostelium discoideum, regulates actin.
Proper regulation of the actin cytoskeleton is essential for cell function and ultimately for survival. Tight control of actin dynamics is required for many cellular processes, including differentiation, proliferation, adhesion, chemotaxis, endocytosis, exocytosis, and multicellular development. Here we describe a putative p21-activated protein kinase, PakD, that regulates the actin cytoskeleto...
متن کاملChemotactic activation of Dictyostelium AGC-family kinases AKT and PKBR1 requires separate but coordinated functions of PDK1 and TORC2.
Protein kinases AKT and PKBR1 of Dictyostelium belong to the AGC protein kinase superfamily. AKT and PKBR1 are phosphorylated at similar sites by phosphoinositide-dependent kinase 1 (PDK1) and TORC2 kinases; however, they have different subcellular localizing domains. AKT has a phosphoinositide 3-kinase (PI3K)/phosphatidylinositol (3,4,5)-trisphosphate [PtdIns(3,4,5)P(3)]-regulated PH (pleckstr...
متن کاملDifferentiation-Inducing Factor-1 and -2 Function also as Modulators for Dictyostelium Chemotaxis
BACKGROUND In the early stages of development of the cellular slime mold Dictyostelium discoideum, chemotaxis toward cAMP plays a pivotal role in organizing discrete cells into a multicellular structure. In this process, a series of signaling molecules, such as G-protein-coupled cell surface receptors for cAMP, phosphatidylinositol metabolites, and cyclic nucleotides, function as the signal tra...
متن کاملRegulation of ammonia homeostasis by the ammonium transporter AmtA in Dictyostelium discoideum.
Ammonia has been shown to function as a morphogen at multiple steps during the development of the cellular slime mold Dictyostelium discoideum; however, it is largely unknown how intracellular ammonia levels are controlled. In the Dictyostelium genome, there are five genes that encode putative ammonium transporters: amtA, amtB, amtC, rhgA, and rhgB. Here, we show that AmtA regulates ammonia hom...
